چکیده

Hypercholesterolemia (HC) is characterized by increased circulating 8-epi-Prostaglandin-F2 (Isoprostane), a vasoconstrictor, marker, and mediator of increased oxidative-stress, whose vascular effects might be augmented in HC. Anesthetized pigs were studied in-vivo with electron-beam computed-tomography after a 12-week normal (n=8) or HC (n=8) diet. Mean arterial pressure (MAP), single-kidney perfusion and glomerular-filtration-rate (GFR) were quantified before and during unilateral intra-renal infusions of U46619 (10ng/kg/min) or Isoprostane (1μg/kg/min). Basal renal perfusion and function were similar, and Isoprostane infusion elevated its systemic levels similarly in normal and HC (333±89 vs. 366±48 pg/mL, respectively, p<0.01 vs. baseline). Both drugs markedly and comparably decreased cortical perfusion and GFR in both groups, while medullary perfusion decreased significantly only in HC. Moreover, MAP increased significantly only in HC (+9±3 and +11±3mmHg, respectively, p≤0.05). Hence, in HC renal functional responses to high-dose Isoprostane are largely similar to normal, but the systemic circulation exhibits augmented sensitivity to pathophysiological levels of Isoprostane and U46619, which may potentially play a role in development of hypertension and vascular injury associated with increased oxidative-stress.

Angiotensin II and Catecholamines Increase Plasma Levels of 8-Epi-Prostaglandin F2 With Different Pressor Dependencies in Rats

We investigated the extent of oxidative stress evoked in the hypertensive rat by measuring plasma levels of 8-epi-prostaglandin F2 (8-epi-PGF2 ), a marker of in vivo oxidative stress. Administration of angiotensin (Ang) II and norepinephrine at doses of 0.7 and 2.8 mg · kg 1 · d , respectively, resulted in similar significant elevations in plasma levels of 8-epi-PGF2 . A 7-day infusion of Ang I...

Altered vascular responses to platelets from hypercholesterolemic humans.

Activated platelets release potent vasoactive factors. Previous studies have focused on mechanisms by which vascular abnormalities lead to altered responses of atherosclerotic arteries. We tested the hypothesis that the activation of platelets from hypercholesterolemic humans produces abnormal vascular responses. Responses to intraluminal and abluminal activation of platelets from normal subjec...

Levels of Eicosanoids (6-oxo-pgf1" and 8-epi-pgf2") in Human and Porcine Lymphatics and Lymph

Prostaglandin (PG)I2 is the primary eicosanoid synthesized by human lymphatics and 8-epi-PGF2", an isoprostane formed during free radical catalyzed peroxidation, is the most potent stimulator of lymphatic contraction tested thus far. We now examine the respective concentrations in the lymphatic wall of both human and porcine lymphatics and lymph fluid using specific immunoassays. Although both ...

Attenuation of endothelium-related relaxation and enhanced responsiveness of vascular smooth muscle to histamine in spastic coronary arterial segments from miniature pigs.

Mechanism of coronary spasm was examined regarding endothelium-related relaxation and contraction produced by smooth muscle cells of spastic vessels isolated from Göttingen miniature pigs. In these pigs, coronary artery spasm was documented angiographically in vivo three months after endothelial denudation, and spastic and control segments of the coronary artery were suspended in organ chambers...

Isoprostane 8-epi-prostaglandin F2 alpha is a potent contractor of human peripheral lymphatics.

Isoprostanes are products of free radical-catalyzed peroxidation and 8-epi-prostaglandin (PG) F2 alpha is the most important vasomodulator of this group of compounds. In human lower leg lymphatics isolated from 5 different patients without a smoking history or hyperlipidemia, 8-epi-PGF2 alpha stimulated in vitro contraction more strongly than the thromboxane receptor agonist U46619. Other isopr...
